DESCRIPTION:Enhancing the efficacy of ADCs in the ovarian cancer tumour micro-environment\nRésumé : \nAujourd’hui encore\, le cancer de l’ovaire (CO) demeure le cancer gynécologique le plus mortel. Malgré un traitement de première ligne efficace associant une chirurgie cytoréductrice et une chimiothérapie\, près de 80 % des patientes rechutent. En effet\, cette tumeur agressive est le plus souvent diagnostiquée à un stade avancé ce qui favorise les récidives et la chimiorésistance\, soulignant la nécessité de nouvelles stratégies thérapeutiques. \nDans ce contexte\, mon équipe a développé un conjugué anticorps-médicament (Antibody–Drug Conjugate\, ADC) ciblant le Receptor for Advanced Glycation End Products (RAGE). Fortement exprimé à la surface des cellules tumorales ovariennes\, ce récepteur participe activement à plusieurs processus clés de la cancérogenèse\, notamment la prolifération et la survie cellulaire ainsi que l’inflammation. Des résultats préliminaires obtenus dans un modèle murin de cancer de l’endomètre ont montré un bénéfice thérapeutique du RAGE-ADC. Son efficacité dans le CO reste à démontrer. \nLe HGSOC est également caractérisé par un microenvironnement tumoral (MET) particulièrement complexe. Celui-ci est constitué de différents compartiments cellulaires\, dont le tissu adipeux. Les adipocytes\, principaux constituants du tissu adipeux\, sécrètent au cours de leur différenciation de nombreux facteurs favorisant la croissance tumorale\, la dissémination métastatique ainsi que l’établissement de la chimiorésistance. Compte tenu de son rôle majeur dans la progression du CO\, il apparaît essentiel de développer des stratégies thérapeutiques ciblant cette composante du MET. La prokinéticine 2 (PROK2)\, ligand de la famille des prokinéticines\, se lie aux deux récepteurs couplés aux protéines G PROKR1 et PROKR2. Cette protéine\, impliquée dans l’inflammation et l’angiogenèse\, inhibe également la différenciation adipocytaire. Par ailleurs\, mon équipe a récemment montré une augmentation de l’expression de PROK2 dans le CO\, suggérant un rôle protecteur potentiel vis-à-vis du développement tumoral. \nL’objectif principal de ma thèse était d’évaluer l’effet du RAGE-ADC sur les cellules tumorales ovariennes en association avec la PROK2\, afin de cibler simultanément lla tumeur et son MET. Ce projet s’est appuyé sur une première caractérisation in vitro de différents modèles cellulaires. Le modèle murin ID8\, utilisé par la suite pour les expériences in vivo\, a été caractérisé afin de confirmer l’expression de RAGE et d’évaluer les effets de l’ADC sur plusieurs processus tumoraux fondamentaux. Cette approche a été complétée par une caractérisation des adipocytes au cours de leur différenciation à partir des cellules 3T3. Une fois ces modèles validés\, un modèle murin immunocompétent de CO a été utilisé afin d’évaluer l’efficacité thérapeutique du RAGE-ADC et de la PROK2\, administrés seuls ou en combinaison. Des expériences complémentaires ont ensuite été réalisées sur des cultures primaires de tissu adipeux murin et humain afin d’étudier les interactions entre les adipocytes et les cellules cancéreuses. Enfin\, les mécanismes moléculaires impliqués ont été explorés par RNA sequencing. Les travaux réalisés au cours de cette thèse ont permis de confirmer l’efficacité thérapeutique du RAGE-ADC dans un modèle murin immunocompétent. Ils ont également mis en évidence\, une synergie entre le RAGE-ADC et la PROK2\, se traduisant à la fois par une inhibition de la différenciation adipocytaire et par un effet direct sur la tumeur. Ces résultats démontrent pour la première fois un effet antitumoral de la PROK2 dans le CO\, renforcé par le RAGE-ADC\, tout en confirmant son inhibition de la maturation adipocytaire. Enfin\, la rédaction d’une revue de la littérature consacrée aux thérapies émergentes dans le CO a permis de souligner l’importance du tissu adipeux dans la progression tumorale et de mettre en évidence l’intérêt de développer davantage de stratégies thérapeutiques le ciblant\, en complément des approches actuellement centrées sur l’immunité. \n_ \nL’accès au CEA est règlementé. DESCRIPTION:We are pleased to announce the 2026 conference « Physics and living systems » of the OLIMPICS federation (CNRS\, UGA\, G-INP\, CEA). It will take place in the Bauges Massif (Lescheraines)\, from Monday\, October 12 at 10:00 am to Thursday\, October 15 at 1:00 pm. \nRegistrations and abstract submissions will be open from now until July 31th. \nThis event is intended to give early-career scientists the opportunity to present their work and build their network. DESCRIPTION:A theory for quantum-critical Planckian strange metal phase in high-Tc cuprate superconductors\nRésumé : \nThe strange metallic state showing incoherent charge transport with universal linear-in-temperature scattering rate: 1/τ = αP kB T / ħ with αP ~ 1 and logarithmic-in-temperature singular specific heat coefficient\, so-called “Planckian metal”\, has been observed in various high-Tc cuprate superconductors over a finite range in doping near optimal doping [1]. Understanding this state is believed to be the key to revealing the mechanism for high-Tc superconductivity. Here\, we propose a generic microscopic mechanism for this state based on quantum-critical local bosonic charge Kondo fluctuations coupled to both spinon and a heavy conduction-electron Fermi surfaces within the heavy-fermion formulated slave-boson t-J model [2]. Our theory is motivated by the striking similarity in strange metal phenomenology between cuprates and heavy-fermion Kondo lattice systems [3]. By a controlled perturbative renormalization group analysis of our effective heavy-fermion model\, we examine the competition between the pseudogap phase\, characterized by Anderson’s Resonating-Valence-Bond spin-liquid made of fermionic spin-singlet spinons\, and the Fermi-liquid state modeled by coherent electron hopping (charge Kondo hybridization). We find an extended quantum-critical metallic phase with a universal Planckian ħω/kBT scaling in scattering rate near a localized-delocalized (pseudogap-to-Fermi liquid) charge-Kondo breakdown transition of the model. The d-wave superconducting ground state emerges near the transition. Unprecedented qualitative and quantitative agreements are reached between our theoretical predictions and various experiments\, including ħω/kBT scaling in optical conductivity [4]\, universal doping-insensitive field-to-temperature (B/T-) scaling in magnetoresistance [5]\, singular specific heat coefficient [6]\, marginal Fermi-liquid spectral function observed in ARPES [7]\, and Fermi surface reconstruction observed in Hall coefficients in various overdoped cuprates [5\,8]. We further applied this framework to obtain a universal quantum-critical B/T-scaling in magneto-transport and unify the B-linear and T-linear Planckian scattering rate observed in LSCO over an extended doping range\, pointing toward a unified quantum–critical origin of Planckian transport in cuprates [9]. This universal Planckian metal phase can be realized within a solvable large-N multi-channel Kondo lattice model generalized from this heavy-fermion formulated tJ model [10]. Our mechanism offers a microscopic understanding of the quantum-critical Planckian metal phase observed in cuprates and its link to the pseudogap\, d-wave superconducting\, and Fermi liquid phases. DESCRIPTION:Structural study of NADPH oxidase complex NOX1 by cryo-electron microscopy\nRésumé : \nMembrane proteins (MPs) constitute a major class of pharmaceutical targets due to their central roles in essential physiological processes. Among them\, NADPH oxidases (NOX) comprise a family of transmembrane enzymes involved in numerous crucial cellular functions — including host defence\, hormone synthesis\, vascular tone regulation\, and equilibrium — through the regulated production of reactive oxygen species (ROS). NOX1\, one of the human NOX isoforms\, is predominantly expressed in the intestinal epithelium\, where it forms a heterodimeric complex with p22phox in the membrane. Dysregulation of NOX1 activity\, often arising from genetic mutations\, has been associated with early-onset colorectal cancer and inflammatory bowel disease (IBD). Elucidating the three-dimensional structures of MPs and their mechanistic properties is essential to accelerate structure-based drug design strategies. Cryo-electron microscopy (cryo-EM) has become a powerful technique for resolving MPs structures at atomic resolutions. However\, structural determination of small MPs (<100 kDa) remains challenging due to their low expression levels\, low signal-to-noise ratios\, dependence on detergent solubilisation\, and intrinsic conformational flexibility\, all which complicating particle alignment and high-resolution structure reconstruction. To overcome these limitations\, antigen-binding fragments (Fabs) are frequently employed as fiducials to increase particle size and stabilise specific conformations by binding to defined epitopes. \nIn this thesis\, a lentiviral-based expression system was developed for the overexpression of the NOX1/p22phox complex in HEK293 T-Rex cells. Initial attempts to determine the structure of wild-type NOX1 were unsuccessful due to low production yield and the small size of the complex\, compounded by the lack of a commercially available monoclonal antibody targeting NOX1. To overcome these limitations\, co-expression with the EROS chaperone\, combined with the engineering of a NOX1 mutant\, named NOX12\, enabled improved production. This mutant incorporates modifications in extracellular loops to allow specific recognition by the anti-NOX2 Fab7D5. This strategy increased particle size\, allowing particle alignment and facilitating structural determination of the complex by cryo-EM. High-resolution structure of the core of NOX12 in complex with p22phox and Fab7D5 was determined at an overall resolution of 2.8 Å. In addition\, the structure of the NOX12/p22phox in complex with EROS and Fab7D5 was resolved at an overall resolution of 3.2 Å. This structure revealed a previously uncharacterised interaction mode between EROS and the catalytic subunit. Furthermore\, the structural framework enabled analysis of disease-associated mutations in NOX1 and provided insights into oxygen access pathways. \nThis work provides the first structural insights into NOX1 isoform in complex with its partner p22phox and together with the EROS chaperone\, establishing a framework for understanding its stabilisation and maturation. In addition\, the successful implementation of protein epitope engineering highlights a broadly applicable strategy for studying other NOX isoforms that lack specific antibodies\, in both resting and activated states\, thereby enabling cryo-EM characterisation across this family of MPs. DESCRIPTION:Evolution du réseau moléculaire associé à LEAFY chez les plantes terrestres\nRésumé : \nApparu chez les algues\, LEAFY (LFY) est un facteur de transcription présent tout au long de l’évolution des plantes terrestres. Initialement connu comme gène essentiel au développement floral chez les plantes à fleurs\, LFY est aussi un régulateur de la formation des méristèmes dans certaines plantes de ce groupe. Chez Arabidopsis thaliana\, LFY agit avec son partenaire UNUSUAL FLORAL ORGAN (UFO)\, qui modifie sa spécificité de liaison à l’ADN et lui permet de réguler des gènes auxquels LFY ne pourrait pas accéder seul. L’interaction entre LFY et UFO est probablement cruciale dans l’histoire évolutive de LFY\, car elle permet une modulation de la spécificité de LFY et élargit ainsi son éventail de gènes cibles. Cependant\, nous ne savons pas quand cette interaction est apparue\, ni si les homologues d’UFO jouent un rôle fonctionnel important chez les plantes terrestres non vasculaires. Notre premier objectif était d’étudier l’évolution de l’interaction LFY-UFO chez les plantes terrestres et son effet sur le réseau de LFY. En combinant des approches phylogénétiques et structurales\, nous avons découvert que les gènes UFO sont restés en faible nombre de copies tout au long de l’évolution et qu’ils seraient apparus avant l’émergence des plantes vasculaires. Afin de comprendre l’évolution moléculaire du complexe LFY-UFO\, nous avons exploré son interaction avec l’ADN chez différentes espèces de plantes terrestres. En dehors des plantes à fleurs\, on ne dispose de données fonctionnelles que pour quelques espèces : la mousse Physcomitrium patens\, la fougère Ceratopteris richardii et les protoplastes du gymnosperme Pinus tabuliformis. Bien que ces données soient précieuses et très informatives\, elles ne concernent que quelques espèces issues de groupes disparates. Ce manque de données nous empêche de comprendre comment LFY a évolué chez les plantes terrestres et comment il a acquis ses fonctions de régulation florale. Notre deuxième objectif était donc de caractériser les fonctions et les cibles de LFY chez les bryophytes Marchantia polymorpha et P. patens. Chez M. polymorpha\, nous montrons que LFY est exprimé dans les structures reproductrices et les méristèmes apicaux\, et qu’il pourrait être impliqué dans l’identité et la différenciation cellulaires. Nous avons exploré comment LFY remplit ses fonctions par le biais de son réseau en aval\, et avons identifié certaines cibles candidates chez M. polymorpha\, tout en démontrant que LFY régule l’expression de l’enzyme LONELY GUY\, impliquée dans la biosynthèse de la cytokinine\, chez P. patens. En replaçant ces nouvelles découvertes dans un cadre évolutif\, nous émettons l’hypothèse que LFY est un régulateur conservé du méristème et de la reproduction sexuée chez les plantes terrestres. L’étude de son réseau chez différentes espèces nous aiderait à déterminer comment la morphologie végétale a évolué d’un point de vue moléculaire.​ \n

DESCRIPTION:Mécanismes régulateurs à l’échelle du génome impliqués dans l’intégration des signaux lumineux et du CO₂ chez Chlamydomonas reinhardtii\nRésumé : \nLes microalgues sont des algues unicellulaires microscopiques que l’on trouve généralement dans les systèmes d’eau douce et marins et qui peuvent vivre à la fois dans les colonnes d’eau et sur le sol. Ces organismes ont développé la capacité de répondre à des stimuli externes potentiellement mortels\, tels qu’une faible disponibilité environnementale de CO2\, au cours desquels ils activent un mécanisme appelé mécanisme de concentration de CO2 (CCM)\, pour augmenter les niveaux de fixation interne du CO2 dans le pyrénoïde de leurs chloroplastes. Ils ont également développé des propriétés photoprotectrices pour pouvoir se protéger des stimuli lumineux excessifs qui pourraient entraîner un stress oxydatif au niveau cellulaire en dissipant les excès. l’énergie lumineuse en chaleur. D’autre part\, les signaux lumineux provenant de l’environnement externe jouent également un rôle crucial dans la photosynthèse et ses mécanismes associés tels que le CCM et la photoprotection susmentionnés. Lorsque nous pensons au processus de photosynthèse et au CO2\, nous pensons principalement au fait que ce dernier est un substrat nécessaire au processus. Cependant\, le CO2 a également un rôle de signalisation et contrôle l’activation transcriptionnelle de la photoprotection et du CCM même en l’absence de lumière. Les mécanismes de contrôle du CO2 sont mal compris car peu étudiés. Dans ce projet\, j’étudie le rôle de signalisation du CO2 et comment il s’intègre aux signaux lumineux au niveau moléculaire chez Chlamydomonas reinhardtii. Je m’intéresse à élucider les mécanismes par lesquels le CO2 et la lumière convergent pour contrôler la photoprotection et le CCM mais aussi l’expression des gènes à l’échelle du génome. Je me concentre dans un premier temps sur le facteur nucléaire CIA5 qui agit à la croisée du CO2 et de la signalisation lumineuse. CIA5\, également connu sous le nom de CCM1\, a été initialement découvert comme régulateur principal des gènes CCM. Les travaux de notre laboratoire ont montré que CIA5 est également un régulateur clé de l’expression des gènes liés à la photoprotection. Au début de cette thèse\, j’ai mis en place des expériences pour commencer à répondre aux questions suivantes : L’expression de quels gènes\, à l’échelle du génome\, est contrôlée/affectée par le CO2 ? Quels gènes sont contrôlés uniquement par le signal lumineux ? Et quels gènes nécessitent que la lumière et le CO2 soient régulés ? Des expériences de séquençage d’ARN ont été réalisées où les algues ont été soumises à différents niveaux de CO2 et disponibilités de lumière. Les données sont analysées avec la direction et l’aide de notre collaborateur\, le Dr Zoran Nikoloski de l’Université de Potsdam. L’objectif est de créer un réseau de régulation génétique (GRN) à partir des résultats du séquençage afin de proposer des gènes candidats contrôlés par le CO2\, la lumière ou les deux. Certains des candidats seront testés en laboratoire tout au long de cette thèse en commandant des souches déficientes en gènes candidats spécifiques respectifs de la bibliothèque Chlamydomonas (bibliothèque CLiP). Les travaux de cette thèse peuvent contribuer à élargir la liste des découvertes qui élucident les rôles de signalisation réels du CO2 et de la lumière chez Chlamydomonas reinhardtii au niveau moléculaire. DESCRIPTION:Modeling the dynamics of DNA (photo)lesions: from reactivity to repair\nRésumé : \nFormation and repair of DNA lesions embrace a rich and combinatorial chemistry\, where atomic-scale simulations are increasingly helpful to complement and extent experimental evidences. \nThe modeling of DNA structure\, dynamics\, and (photo)chemistry has benefited from a series of recent methodological developments that now allow\, for instance capturing DNA-photosensitizers interactions\, probing new excited-state mechanisms for DNA lesions induction or photostability [1]\, and rationalize the photochemistry or photocatalytic properties of drugs within DNA owing to hybrid QM/MM-MD schemes. \nI will present a series of examples where computational approaches can palliate\, at least partly\, the absence of NMR\, FRET or X-ray data for damaged DNA oligonucleotides\, DNA-drug\nbinding modes or even DNA-proteins interactions at the nucleosomal scale [2\,3]. DESCRIPTION:Superconducting Epitaxial Silicon – when you turn on the light\nRésumé : \nSince the discovery of BCS superconductivity in silicon by nanosecond laser ultra-doping with boron\, theoretical and experimental works have endeavored to understand what triggers and controls the superconducting phase. Indeed\, superconducting Si has great potential to develop a cryogenic electronics with the advantages of large scale integration and high reproducibility. Through the optimization of the nanosecond laser temporal profile\, we achieved an excellent control of both the electrical and structural properties of ultra-doped Si thin layers\, with state of the art maximum carrier concentration (8 at.%) in monocrystalline epilayers with few defects\, 100% dopant activation up to and above the solubility limit\, and a vertically homogeneous doping profile [1-3]. The control and improvement of the active doping is directly reflected in the control of the superconducting critical temperature Tc\, increased by 30% up to 0.9 K in this optimized setup\, and whose dependence with doping can now be modelled within BCS frame [3]. Furthermore\, we demonstrated that superconductivity is not only controlled by doping\, but also by the lattice deformation. Thus\, it is possible to tune up to 50% Tc by modifying by 1% the lattice parameter\, as shown through nanosecond laser incorporation of Ge up to 20 at.% [4]. \nThe sensitivity of superconducting silicon allows for multiple modulation ‘knobs’: for instance Tc can be tuned by doping\, allowing for the design of superconducting\, metallic or semiconducting regions in a device\, or by strain relaxation\, e.g. through localized Ge ion implantation. In addition\, the thin superconducting silicon layer can be affected by the absorption of photons either in the superconductor itself\, or in the undoped silicon underneath. The effect of light irradiation was highlighted in superconducting coplanar waveguide resonators\, whose resistivity easily matches the vacuum impedance. Due to the large disorder\, a high kinetic inductance characterizes the resonators [5]. The quasiparticle dynamics was measured when submitting the resonators to either a light or a microwave pulse\, these out-of-equilibrium measurements being also compared with the equilibrium generation-recombination noise spectral density. In addition\, work is in progress to assess the effect of visible photons on the DC response of the superconducting silicon micrometer-size lines\, where a single-photon counting mode with extremely low Dark Count Rates has been highlighted\, and whose detection mechanism is currently investigated. \n1. DESCRIPTION:Supramolecular Nanocages as Masks for Fullerene Regiofunctionalization and Beyond\nRésumé : \nThe design of a confined cavity dictates the type of guest to be encapsulated\, and supramolecular cages are tunable scaffolds that allow the rational design of their cavities. Nowadays\, easily accessible C60 and C70 fullerene mono-adducts are mainly used in any application (1) due to the hampered accessibility to pure alternative fullerene poly-adduct derivatives. In general\, multi-adduct mixtures with uncontrolled regioselectivity (multi-isomers) are obtained\, and chromatographic purification is too costly and time-consuming. Herein\, porphyrin-based supramolecular nanocapsules (2\,3) are used as supramolecular shadow masks to tame the over-reactivity of Bingel-type cyclopropanation reactions and to have full control over the equatorial regioselectivity\nand the number of additions. Furthermore\, the regioselectivity control is finely tuned using a three-shell Matryoshka-like assembly towards synthesizing a single trans-3 bis-Bingel-C60 for the first time (4). Also\, the mask strategy is extended to C60 and C70 for Bingel and Diels-Alder (5\,6). We envision that the described protocol will produce a plethora of derivatives for applications such as solar cells. DESCRIPTION:Towards confined gold catalysis\nRésumé : \nGold complexes have been the subject of an intense research over the past decade.[1] The main driving force to this interest is to impart new reactivity to these complexes\, that can be leveraged in catalysis. In addition to the widely used ligand engineering strategy\,[2] the concept of confining gold complexes within supramolecular cages has also shown a great\npotential.[3] Toste et coll. have demonstrated\, that small cationic gold complexes (LAu+\, L = Me3P)\, generated within anionic cages show a markedly different reactivity compared to their non-confined analogues. However\, the small cavity size of the cage (251 Å3) has not allowed to extend this concept to larger complexes (L = tBuP\, NHC) and substrates. Indeed\, one obstacle\nto overcome is the synthesis of large anionic cages\, that is far from being straightforward.\nIn order to design and synthesize large anionic cages\, we have implemented a rational approach combining modellizations and cavity size calculations.[4] As a result\, we have prepared an anionic\, cyclotricatechylene based supramolecular cage 1 with a large cavity (558 Å3).[5] Recently\, we have been studying the host-guest chemistry of this cage\, in order to prepare the confined gold-catalyst (Au+@1\, Figure 1). The catalytic activity of Au+@1 is being studied\, with special attention to the synthetically challenging gold catalyzed macrocyclizations. \nFigure 1. DESCRIPTION:Synthèse et caractérisation de polymères d’accepteurs non fullerène stables pour des cellules solaires « tout polymère »\nRésumé : \nAu cours des cinq dernières années\, les cellules solaires organiques (CSO) ont montré une forte progression de leurs performances dépassant 20% de rendement photovoltaïque en 2022. La formulation de ces deux matériaux sous forme d’encre imprimable rend possible l’industrialisation de ce procédé sur des substrats flexibles et par des techniques d’impression à haute vitesse. La bascule vers l’industrialisation à grande échelle est proche mais il reste à s’affranchir de plusieurs verrous technologiques importants. Parmi les approches permettant d’augmenter la stabilité des couches actives\, le remplacement des petites molécules acceptrices appelée accepteurs non fullerène par leurs dérivés polymères a récemment attiré notre attention. Ils présentent plusieurs avantages : une grande facilité de formulation sous forme d’encres solubles dans des solvants non halogénés\, une nano-morphologie robuste permettant une stabilité à long terme et l’accession a des couches plus épaisses grâce au mélange de deux polymères.​​​​​

DESCRIPTION:Dynamic Supramolecular Eutectogels with Reversible Light Driven Reconfiguration\nRésumé : \nDeep eutectic solvents (DESs) constitute a highly tunable class of liquid media formed by the association of two or more components (e.g.\, a hydrogen-bond donor and an organic salt)\, whose extensive hydrogen bonding interactions result in a pronounced depression of the melting point. Their composition-dependent physicochemical properties make DESs particularly attractive for directing the self-assembly of functional soft materials [1\,2]. In this context\, eutectogels (gels formed within DES media)\, offer a versatile platform in which solvent–solute interactions can be exploited to modulate macroscopic material properties beyond those dictated by the gelator alone [3]. \nHerein\, we report a versatile strategy for the design of light-responsive eutectogels with intrinsically tunable mechanical properties. The approach relies on the supramolecular co-assembly of an amphiphilic component with a rationally designed azobenzene derivative engineered for high solubility in water-free DES media. Through complementary characterization across molecular and mesoscale length scales\, we provided a detailed understanding of the heterotypic interactions underlying the formation of elongated\, one-dimensional supramolecular networks. We further establish that photoisomerization of the azobenzene units induces pronounced rearrangements of these networks\, providing a molecular-level mechanism for controlling the material’s hierarchical organization. \nUpon ultraviolet irradiation\, this molecular reconfiguration leads to an abrupt gel-to-sol transition\, effectively erasing the material’s solid-like mechanical response. Subsequent exposure to visible light reverses the process\, restoring the gel state and recovering a mechanically robust network that remains stable during prolonged storage without appreciable loss of performance. Remarkably\, the optically driven transition is highly reversible and cyclable\, enabling repeated switching between gel and sol states without detectable deterioration of structural integrity or mechanical properties. DESCRIPTION:Etude des fonctions catalytiques et non-catalytiques de la déubiquitinase USP36 chez Drosophila melanogaster\nRésumé : \nL’ubiquitination est une modification post-traductionnelle essentielle à la régulation de nombreux processus cellulaires tels que la croissance\, la prolifération\, la transcription\, la réparation de l’ADN ou encore l’apoptose. Cette modification est réversible grâce à l’action des déubiquitinases (DUB)\, des enzymes capables de retirer les molécules d’ubiquitine des protéines cibles et ainsi de moduler leur stabilité\, leur localisation et leur activité. Parmi elles\, USP36 (« Ubiquitin-Specific Protease 36») est une déubiquitinase nucléolaire impliquée dans la régulation de la stabilité de plusieurs protéines\, dont le facteur de transcription c-MYC\, un régulateur majeur de la croissance cellulaire et un oncogène fréquemment dérégulé dans de nombreux cancers.\nCette thèse vise à caractériser les fonctions catalytiques et non catalytiques de USP36 in vivo\, en utilisant Drosophila melanogaster comme organisme modèle. Pour distinguer les fonctions dépendantes de son activité enzymatique de celles liées à la présence de la protéine elle-même\, une lignée mutante exprimant une forme de USP36 dépourvue d’activité catalytique a été générée. L’étude de ce modèle révèle que\, contrairement à l’absence complète de USP36\, la perte de son activité catalytique n’empêche pas le développement jusqu’à l’âge adulte. Ces résultats mettent ainsi en évidence des fonctions de USP36 indépendantes de son activité de déubiquitination\, notamment dans le contrôle de la croissance et de la prolifération cellulaire. En revanche\, l’activité catalytique de USP36 s’avère essentielle à la fertilité mâle. Les mutants catalytiques présentent une diminution importante de la production de spermatozoïdes matures\, associée à des défauts de spermatogenèse. L’analyse de la spermatogenèse met notamment en évidence des altérations de la différentiation et de l’individualisation des spermatides\, révélant un rôle spécifique de l’activité enzymatique de USP36 dans le développement des cellules germinales mâles. Des analyses complémentaires suggèrent une implication de USP36 au niveau de structures nucléaires spécifiques des spermatocytes appelés Y-loops\, avec un impact sur la transcription des gènes portés par le chromosome Y.\n​ Au final\, ces travaux montrent que la présence de la protéine USP36\, même dépourvue d’activité catalytique\, est suffisante pour assurer certaines fonctions essentielles à la croissance et à la prolifération cellulaire\, et que son activité de déubiquitination est spécifiquement requise pour la spermatogenèse. Cette étude révèle ainsi une dissociation fonctionnelle entre les activités catalytiques et non catalytiques de USP36 in vivo et souligne l’importance de prendre en compte et de distinguer ces différentes lors de l’étude des déubiquitinases dans leur contexte physiologique ou pathologique. ​​​​

DESCRIPTION:Etude de l’impact de l’inhibition pharmacologique de l’activité enzymatique de la déubiquitinase UCH-L1 sur la signalisation calcique\, les canaux ioniques et le protéome des neurones corticaux de souris\nRésumé : \nL’ubiquitination est une modification post-traductionnelle qui vise à la liaison covalente de l’ubiquitine sur des protéines cibles. Ce mécanisme contrôle la destinée des protéines\, via leur dégradation par le protéasome ou les lysosomes\, et régule de nombreux processus biologiques comme l’endocytose\, la localisation subcellulaire ou l’activité des protéines ubiquitinées. Elle est réversible grâce à des protéases appelées déubiquitinases (DUBs) dont UCH-L1\, qui est prédominante dans le cerveau. Cette enzyme a été associée à plusieurs maladies neurodégénératives mais ses fonctions biologiques et ses substrats restent encore partiellement caractérisés. Afin d’approfondir la compréhension de ses rôles cellulaires\, nous avons utilisé deux inhibiteurs pharmacologiques sélectifs de UCH-L1 : l’IMP1710 et le GK13S.\nDes expériences d’imagerie calcique Fura-2 montrent que l’inhibition de UCH-L1 réduit l’influx de calcium (Ca2+) à travers les canaux calciques dépendants du voltage (VGCCs) dans des neurones corticaux d’embryons de souris\, principalement via les canaux sensibles aux dihydropyridines (VGCCs de type L). Ces inhibiteurs réduisent également la quantité totale de protéines Cav1.2 dans les neurones mais pas dans les lignées cellulaires cancéreuses. Des expériences de biotinylation de surface ont montré une réduction de la quantité de protéines Cav1.2 à la membrane plasmique. De plus\, l’inhibition de UCH-L1 diminue la taille des réserves de Ca2+ mobilisables du réticulum endoplasmique et des lysosomes\, ainsi que l’entrée capacitative de Ca2+ (store-operated Ca²⁺ entry). Nous avons ensuite établi que l’inhibition de UCH-L1 réduit d’autres flux ioniques dont ceux à travers les récepteurs NMDA et les canaux TRPC6 et TRPM7.\nEn parallèle\, une double approche protéomique quantitative par spectrométrie de masse (marquage isobare et sans marquage) a été réalisée afin de caractériser les potentiels substrats de UCH-L1. Cette stratégie a permis d’identifier sept protéines communes aux deux techniques dont l’abondance diminue fortement suite à l’inhibition de UCH-L1. Parmi celles-ci figure USP11\, une DUB connue pour son implication dans le développement du cortex cérébral. Ce résultat a été confirmé par western blot dans les neurones\, sans effet équivalent dans les cellules cancéreuses.\nEn conclusion\, cette étude met en avant le rôle de UCH-L1\, l’une des principales DUBs cérébrales\, dans la régulation du transport ionique à travers la membrane plasmique des neurones et identifie USP11 comme l’un de ses substrats potentiels. Ces travaux apportent des connaissances nouvelles sur les fonctions cellulaires de UCH-L1 et contribuent à approfondir notre compréhension de son rôle dans le cerveau . ​​​​

DESCRIPTION:We are pleased to announce that pre-registation for the SFN School on TOF spectroscopy is now open. The school will take place 7-10th of December 2026 at the Institut Laue-Langevin in Grenoble.\n\nThe objective of this school is to accompany the French neutron community towards the use of Time-of-flight spectrometers in spallation sources\, and especially in preparation for the use of the upcoming instruments suite at the ESS.  This school is addressed to participants (permanent and non-permanent) who are already familiar with neutron spectroscopy.
