SUMMARY:Soutenance de thèse par Alessio D’ACAPITO (CEA-Irig/IBS)
DESCRIPTION:Study of E. Coli cell wall perforation by bacteriophage T5\nRésumé : \nBacteriophages are fascinating nanomachines that are highly specific and remarkably efficient in infecting their bacterial hosts. While the cell envelope perforation mechanisms by myophages\, such as T4 or of podophages\, such as P22 has been well characterized\, less is known about the molecular mechanisms of the infection of siphophages\, such as T5. \nIn this work\, we used an integrative approach combining cryo-electron tomography (cryo-ET)\, subtomogram averaging (STA) and single particle analysis (SPA) to obtain snapshots at successive stages of phage T5 infection\, from early cell recognition to post DNA injection state. We used E. coli minicells as a simplified cellular model and developed a tailored grid preparation protocol to control the timings of infection and focus on specific timepoints. \nFirst\, we solved the structure of T5 L-shaped fibers at 4Å resolution by SPA. Second\, we characterized by SPA the structure of the adsorbed but noninfecting phage at 4°C by describing a new intermediate of the tube anchoring mechanism\, where the tail tube is brought close to the membrane by the bending of the central fiber but the tube is still closed ; we could conclude that the Cterminal part of the tape measure protein (TMP) pb2-C plays a key structural role. \nLastly\, we solved by STA the structure of the infecting phage tip docked on the minicell and of the trans-periplasmic channel that allows the DNA to be translocated into the bacterial cytosol. We propose a model for the channel\, formed of three copies of the TMP. \nBeyond structural biology\, my PhD work was also focused on building tools for the cryo-ET local community. I have developed a tool for “erasing” the membrane signal from sub-volumes extracted with Dynamo as well as a beginner-friendly method for quick tomogram reconstruction from raw data. We made both publicly available on GitHub.

SUMMARY:Valeria DEL CAMPO (Physics Department\, Universidad Técnica Federico Santa María\, Chile)
DESCRIPTION:Graphene in photovoltaic solar energy : results and challenges\nRésumé : \nGraphene has attracted significant interest in photovoltaic applications due to its unique combination of high electrical conductivity\, optical transparency\, mechanical flexibility\, and chemical stability. Despite these advantages\, several scientific and technological challenges still limit its integration into high-performance solar cells. This seminar presents recent results from our research group on the use of graphene in different photovoltaic technologies\, with particular emphasis on the role of interfaces and surface electronic properties. \nFirst\, the implementation of graphene as a transparent conductive electrode in silicon heterojunction (SHJ) solar cells will be discussed. In this context\, graphene offers a promising alternative to conventional transparent conductive oxides\, while enabling new device architectures. The second part of the talk will focus on the use of graphene as an encapsulation layer to stabilize copper oxide hole-transport layers in perovskite solar cells. While graphene can provide effective protection against degradation processes\, the subsequent deposition of high-quality perovskite films onto graphene-covered surfaces presents significant difficulties due to the surface chemistry and wetting properties of graphene. \nFinally\, the seminar will address the characterization of surface electronic properties through work function measurements and inverse photoemission spectroscopy. By probing the electronic structure and energetics of graphene-based interfaces\, these techniques contribute to a deeper understanding of charge transport mechanisms\, which remain among the main challenges limiting the implementation of graphene in high-efficiency photovoltaic devices. \n  \nShort Bio/CV \nDr. Valeria del Campo is an Associate Professor in the Physics Department at Universidad Técnica Federico Santa María (Chile)\, where she currently serves as Head of the Engineering Physics Program. She coordinates the Solar Sustainable Solutions research line at the Solar Energy Research Center (SERC Chile)\, a multidisciplinary center dedicated to advance in public policies based on scientific findings. \nHer research interests focus on photovoltaic solar energy\, graphene and other advanced materials for solar cells\, and the impact of environmental factors such as dust deposition on photovoltaic performance. Her work combines surface and interface characterization with the development of materials and technologies for high-efficiency and sustainable solar energy conversion. \n_ \nContact : deborah.verger@grenoble-inp.fr \n

SUMMARY:Soutenance de thèse par Simon PAUL (CEA-Irig/BGE)
\n_ \nContact : irig.communication@cea.fr

SUMMARY:Soutenance de thèse par Francesca MOSCA (CEA-Irig/IBS)
DESCRIPTION:Targeting L\,D-Transpeptidases in Lpp-Peptidoglycan Crosslinking in E.coli\nRésumé : \nThis thesis investigated the structure and function of L\,D-transpeptidases (LDTs) responsible for covalently attaching Braun’s lipoprotein (Lpp) to the peptidoglycan layer in Gram-negative bacteria. Because Lpp constitutes the principal covalent connection between the peptidoglycan and the outer membrane\, disruption of this linkage destabilises the bacterial cell envelope. Understanding the molecular mechanisms underlying LDT-mediated Lpp attachment therefore provides a foundation for developing novel inhibitors capable of compromising outer-membrane integrity and enhancing the activity of existing antibiotics against antibiotic-resistant Gram-negative bacteria. The work focused on mainly two aspects of LDT function : the interactions of these enzymes with existing and novel antibiotics\, and with their physiological substrates Lpp and peptidoglycan. Particular emphasis was placed on LdtB\, the most essential LDT in E. coli. Structural and functional characterization was performed using NMR spectroscopy\, biophysical methods\, and biochemical techniques\, complemented by in silico approaches\, including molecular dynamics simulations. This integrative approach was used to decipher the molecular determinants of Lpp-peptidoglycan transpeptidation mechanism and to propose a possible stuctural model for the reaction\, which remains poorly understood despite its biological relevance. In this context\, E. coli served as a model system\, whose study may provide insights into mechanisms that could be conserved across other Gram-negative bacteria. This work was carried out within the framework of the BREAKthrough consortium\, which enabled part of the research to be conducted in collaboration with partner laboratories across Europe.
